When you have a multi-instrument piece, is there a way to play back one instrument from the score rather than having to extract the instrument?

I don't know PrintMusic, but in the full Finale you can use the Instrument List to Solo or Mute individual instruments:
Window menu > Instrument List...

The same in PM. For a quick listen, you can hold down shift and spacebar, and click in the desired staff.
