I have a repeat section with multiple instrument. I want one instrument to play only on the repeat, not the first time through. Is there a way to do that?

You are talking about playback, not about layout, right?

More info needed.

Take a look in the MIDI/Audio menu.
In the top of the menu, what option is selected?
Is it Play Finale Through MIDI? - or VST?

The general solution is to use the Expression Tool.
Add an expression that mutes the instrument on one Pass, and add another expression that un-mutes the instrument on the other Pass.
On which Pass the expression “does its thing”, is defined in the expression’s chiclet for Playback.

1) With playback through MIDI you can use Controller 11.
To mute, let the expression change Controller 11 to zero.
To un-mute, let the expression change Controller 11 to 127.

2) With VST playback the method is similar.
To mute, change the instrument to an un-used channel.
Then, change it back.
